Gill came off the bench in Osaka seven days ago to earn his fourth cap and has been called into the starting XV by head coach Robin McBryde for the second test.
The 26-year-old, who helped Wales to win the Grand Slam in 2012, will make his first start since the Barbarians fixture in Cardiff last June and is looking to take his opportunity.
“It’s really good to be back in the mix,” said Gill.
“It’s been a while, it’s my first start since the Barbarians game last year and I’m hoping it will go well and I can make a mark and make a statement.
“I was happy with the way I went last week from the bench, I tried to bring some energy on the pitch and hopefully I did that.
“Hopefully this weekend we can put a performance together for 80 minutes and show what we are able to do.”
The Wales squad trained against Japanese club champions Suntory Sungoliath on Tuesday in Tokyo and Gill believes that experience will benefit the squad ahead of the weekend.
“It was a really good session and they got stuck into us,” he added.
“They are a very well drilled team and have amazing facility and it was good for us to go against them.
“We have got a really competitive squad, obviously its friendly rivalry but there are a few of us all fighting for the same position so it really helps to raise the game.”
